Ms. Cow is named after Fred's aunt, Harriett Elizabeth Rogers. He mentions this in the Archive of American TV Interview

I grew up watching & loving Mister Rogers, and it always seemed like Harriet Cow was featured more prominently in the pre-1975 episodes. We saw her as the teacher some in the post-1979 shows, but rarely saw her with Donkie Hodie again like before. Once the school was built, she was usually doing that instead of working in the potato harvesting with Donkie.
